Retatrutide is a 39 amino acid peptide that functions as a triple agonist, stimulating the glucagon receptor, glucose-dependent insulinotropic polypeptide receptor, and glucagon-like peptide-1 receptor. It is a modified GIP peptide with non-coded amino acids like Aib2 (α-amino isobutyric acid) at positions 2 and 20, which enhance stability and contribute to GIP activity. It also includes an α-methyl-L-leucine (αMeL13) at position 20, which also contributes to GIP and glucagon activity.
